I've also found there is a mild inconsistency between the color at the top and the bottom of the screen, but it's hardly noticeable unless you a hardened CRT user and photo/graphics editor like me.
Don't know about your dark spot issue, but the above color issue is something I noticed from day one of having my Flat Panel iMac. I've been told it's perfectly normal for LCD screens.
