I run a free e-mail service on our web site and currently we have @4000
users(in less than 2 months!) and are increasing by @100 per week. We're
running a P-III 500 with 512MB of RAM, and 2 9GB UW SCSI Drives and it is
VERY content with IMail and IIS4 running alongside of each other(and SQL
Server 7). We get @2 million hits per week and this box is more than
capable of handling the load. I figured I would share what we run to give
an idea ;-)

The largest system I've heard about was around 200-220K users on a single
PC.
This was well over a year ago, so todays best hardware should be more than
enough to support a large user base. I do not recall all the details of your
proposed system, but a very fast CPU, lots of RAM and a large and fast disk
array are all essential to max performance of IMail.
IMail has a 'Peer' capability that allows a single domain to span several
computers. The only drawback to this is that users are assigned to a
particular host and that host is the only one that has their mailbox. The
must
connect to that host to read their mail.
With a large system, you might also consider the need for a backup mail
server. And Backup of the data that the mail system stores. A single system
could mean a single point of failure could down your mail services. I'd hate
to have that many users on my back, especially for a minor single point
failure!
