Since many/most spam MAIL FROM's are spoofed, it would seem dangerous to use the kill.lst file to deny spamming servers from connecting your iMail machine based on the MAIL FROM contents... so this kill.lst file is useless in an automated/scripted context, correct?

The Kill List is designed for blocking E-mail from return addresses that you do not want. For example, if the New York times refuses to stop spamming you, you can block @nytimes.com. However, as you note, it is not designed to automatically block E-mail addresses that send spam.

What would be cool is if kill.lst or some other text-based file, say "killIP.lst" could contain a list of IPs that should be killed, and respond with a similar error like "501 blacklisted source IP".

Any way to create such a situation in iMail?

No. The SMTP Control Access file will block the IP, but without any information on why it is being blocked. And, it isn't in a text file.
