For example start WordPad and write mailto:[email protected]. This should give a clickable text. Click on it. Does Thunderbird open? Or look into the registry in HKEY_LOCAL_MACHINE\SOFTWARE\Clients\Mail whether Thunderbird is listed with the correct DLLPath.
