You are asking about spot colors in the title of the message, but then talking about (and exampling) process colors.
Which are you interested in??
If I have two spot colors defined, is it possible to set the fill color to a combination of the both of them?
In PDF, yes - it's called DeviceN. However, I don't believe iText supports it currently.
