No, just one.
Put it in .exrc if it should apply to straight vi, as well as vim and
gvim. Put it in .vimrc if it shouldn't apply to straight vi, but should
apply to vim and gvim, etc.
I suspect that "set mouse" is a vimism, and as such should go in .vimrc.

I searched for .vimrc and found /etc/vimrc. I had a few problems after
adding that line there. Then I discovered my error. (I had to use
something other than vi to fix it.) Then since I could find no .vimrc
anywhere, I *assumed* I should create it in my home directory and add
*only* that line. Viola!
