Use the Relationship Calculator Tools > Relationship Calculator You can have the Mayflower Ancestor on the left and your niece on the right. Select the relationship you want (if there is more than one because of cross marriages they will all be shown). Use the Print Button then select print current relationship to create a chart showing only the direct line ancestors between your niece and the Mayflower immigrant.
Warning, if there are more than one relationship to NOT use the print all, there is a problem with that option right now and the charts will not be correct.
