The line doesn't compile, and spits out an error. It looks like you
meant to write
\override BreathingSign #'text = \markup { \fontsize #8 { \musicglyph
> #"comma" } }
As Shane said, it's much cleaner to override the font size as in the
following snippet.
\new Staff \relative c' {
\override BreathingSign #'font-size = #3
c1\breathe
}
