Quad speed CD-ROM data rates are around 600 k/s, so I would get as close to that as you can while balancing quality. I personally don't worry about data rate too much; I just go by the spatial quality of the compression. You can generally compress Sorenson down to 50-60% without much noticeable quality loss. I also recommend not going too much over 320x240 for CD-ROM use. if you want it bigger, try using Sorenson and compress it at half size, then display it at double size. Sorenson resizes excellently, and it saves you a butt-load of processor speed.

Consider putting a stretched still image at the start of your Quicktime movie in a really easy-to-load format - e.g. something which uses run-length-encoding.
