At the prompt, type >/var/log/wtmp
that will truncate the file.  The only slightly bad effect is that the
output of the w command (and other similar ones) will be wrong until all
users log out.

Basically, yeah.

You might want to look at the logrotate program which will automate a lot
of this for you.
