I think it does make a difference... especially when cleaning up things like the 'lap lines' on pixel at a time, when you join multiple scans together into a poster-sized print image, that his going to be printed at a very high dpi number, and then (hopefully) be sold to a customer. It might also matter if you are watching DVD movies.
