Have you tried to connect with the sa username?  Are you sure that the user you are trying to connect with is associated with the db you are trying to connect to?
 
You can't create an ODBC datasource outside of CFMX...As you know the JDBC datasources must be set up in the CFMX admin. 
 
I have connected with SQL Server without an issue....I did use the sa login with a password....Strange you can't connect.

For the life of me, I can not get ColdFusion MX to create a new datasource or recognize a current datasource.  The ODBC system datasources on the server work fine in connecting and using the sqlserver username and password with admin rights.  But when I try to create and verify those exact same datasources on the CF Administration screens I get a login error.  Initially I received a generic can’t connect error, then I installed the new updater and now I receive a login error.  Does it have anything to do with the JDBC drivers?

The root cause was that: java.sql.SQLException: SQLException while attempting to connect: java.sql.SQLException: [Macromedia][SQLServer JDBC Driver][SQLServer]Login failed for user 'aUser'.
