I ran into this recently as well. One of the issues I had was in setting up a SQL2K connection on another machine. Using the JDBC drivers for MS SQL Server you connect via TCP/IP, thus I had to open port 1433 for the CF to talk to the SQL box. Another issue is the SQL box was not set to use TCP/IP since someone turned this off during the initial install.
 
Also, when I used the ODBC Socket selection within CF I connected to my previously created SQL SystemDSN I without a hitch. I am not too sure which connection is faster, but for some reason a little voice in my head said to use the MS SQL Server connection option.
 
Finally, I know you would have checked this, but does the user account you are using to access the db have the proper privileges?

For the life of me, I can not get ColdFusion MX to create a new datasource or recognize a current datasource.  The ODBC system datasources on the server work fine in connecting and using the sqlserver username and password with admin rights.  But when I try to create and verify those exact same datasources on the CF Administration screens I get a login error.  Initially I received a generic can’t connect error, then I installed the new updater and now I receive a login error.  Does it have anything to do with the JDBC drivers?

The root cause was that: java.sql.SQLException: SQLException while attempting to connect: java.sql.SQLException: [Macromedia][SQLServer JDBC Driver][SQLServer]Login failed for user 'aUser'.
