There have been several 2wd conventional MCs built. Yamaha had an experimental 
fluid drive, more than one company have
tried mechanical drives, usually down one fork leg. Knowledgeable folks have 
told me the traction out of a sharp turn,
where you would heavily weight the front wheel, is great. I question the rest 
of the utility because you so often want
the front wheel light. Perhaps on a downhill, having the front driven would add 
stability? In sand it might be great
too. To my knowledge, no one has done a road racer.

Now a three or four wheeler might make more sense with hub motors. You still 
have the complexity, overall weight, and
unsprung weight problems though. Theres just not that much wrong with 
drive-shafts and U-joints in conventional
vehicles. In bikes I still say the front-drive is not useful, but then I've 
never ridden a 2wd bike.
