I've created the Workflow using Sharepoint Designer, I'm not sure if I can perform the following with it.

To access what that currently is, you can write a console app, and use the object model to inspect SPWorkflowAssociation.AssociationData which will provide you with the XML you're currently passing. You can then either update/add the InitLock property to 'true', that should do it! Apparently if you pass <my:InitLock>true</my:InitLock> as part of the initialisation data, you will get the desired affect.
