Not really. The easiest way is to look at all the bugtrackers, investigate the bugs, confirm or not, and eventually propose patches for them. Finally, for the knowledge, well, it depends on the part you are working on. LXDE is mostly C + GTK, some part of Lubuntu are python (LSC for example).
