I had this problem installing 1.4.5 on my windows machine.

Try erasing the preferences directory located at:

c:\Documents and Settings\*username*\Application Data\lyx1.4.x

This is to regenerate the directory in full. It will work at least the first time

If this does not work, double check your tex installation. If you are running Miktex 2.5, download it to your computer before installing it and do not use the Lyx installer (I found some problems of this kind, using it)

Now my system is up and running with Lyx 1.3.5 and Miktex 2.5

Is this on a Windows system? If so, is your home directory on a remote server? (If you are not sure, open a DOS window, run 'set h', and see what is set for your home drive and home path.)
