There is accessibility settings to turn on a voice for the Kindle.
I did it once for a client, but I did not spend any time to determine if I 
could access all the menus.

No, Kindle books are not accessible on the Mac. There might be a way to jump 
through a bunch of hoops and lose the drm and thus be able to access them. 
but if you're talking about normal regular access that sighted people take 
for granted, no. Can't do it.
