Yes - create a new image and turn on encryption. While TrueCrypt is a great 
suggestion too, I'm partial to tools that are built-in, all other things being 
equal. Granted, TC does a lot that encrypted Disk Images won't - but if you 
just need an encrypted filesystem, stored on a read-only medium, DMGs are nice 
and easy.
