First: Make sure Mailplane is your default email application:
- Open Preferences
- Click 'Advanced'
- Select 'Mailplane' as your default email application.


This will allow Mailplane to handle 'mailto' links.
As your web browser already handles them in another way: Which web browser do 
you use? Did you install any extension that opens Gmail?
