I prefer Growl as it's practically infinitely customizable. OS X notifications are not. Since I use 2 monitors, OS X notifications often get missed as they display on the main monitor. With growl, I can tell the notifications where, when, how long and what to look like. I can customize them for each app as well so I know what app is notifying me right off the bat.
And as for signatures - I use multiple signatures and Gmail only allows 1 sig file per email address. Totally doesn't work for me. So, one of the features I've always loved most about Mailplane is the customizable sig files and the ability to have multiple files for each email address. So much less complicated. I've had many email conversations with Ruben throughout the Mailplane 3 beta about the sig files so I'm very disappointed to find they're not available.
