Hi, While I was playing with rendering OSM data from Spatialite database with Mapserver I found an issue that is reported in http://trac.osgeo.org/gdal/ticket/4212 Perhaps it is worth having a look if Mapnik SQLite plugin already is using the Spatialite spatial index table as it should.
