Ok, well I'm going to ask a potentially stupid question.  On your SMTP server, 
for us when we need to add a new server to be allowed to use the smtp ...we 
have to go into  IIS 6.0 manager, the SMTP virtual server properties, and under 
Access, Connection (and also Relay), add in the new server IP address.  That's 
so that only our defined processes can use that particular SMTP service.  Did 
you confirm that this new server 2012 r2 box has the access rights to that smtp 
service ?
