It'd be the same for any application you want to start when the system
boots: put a desktop file in /etc/xdg/autostart.  For your particular
needs you could have the desktop file launch a script that launches
xinput_calibrator and then removes the desktop file from the autostart
directory so that on subsequent boots, it won't launch.

A better solution may be to get the calibration values ahead and create
an entry in /etc/X11/xorg.conf.d/* for your specific touchscreen.  That
way it just works and no calibration would be needed.
