If the problem is a fuel injector not working at all, no. If it is an ignition problem, maybe. A cylinder not firing will cause fuel to wash down that cylinder and contaminate the oil in the crankcase. It is probably best not to drive it.

If you can identify the culprit, unplug and/or cap off the injector.
No fuel at all that way, it should be fine.  With no emissions package
to get PO'd about the extra air there should be no harm done.

If the miss is bad enough maybe you could use a thermometer of some
sort to find the bad cylinder by looking at the temperatures of
each leg of the exhaust manifold?  I found a dead hole on the
Hercules that way.
