In addition to establishing the linear range of your film using a
standard curve as Wo suggests, be sure that your bands are not
saturated. Everything should be within the grayscale, no true black (for
black bands on white background) or white (for white bands on black
background).

To convert the data that you will get into meaningful ones, you'll
probably need a standard curve within the actual picture. A problem
you might run into is the very low dynamic range of xray film and
thermal printouts. If you have the possibility, use fluorescence
scanners (e.g. Licor Odyssey) and cameras instead.
