It depends on the autolearn settings in your local.cf or sa-mimedefang.cf. (man Mail::SpamAssassin::Conf for details)
If you've got Bayes running under MIMEDefang already, you're more than halfway there.
If you do end up training manually, make sure local.cf has the same sitewide bayes settings as sa-mimedefang.cf. (Or just run it as the defang user, depending on how you have it set up.) You want to make sure you're training the same database you'll be using!
