Could it be a transient startup problem? How about permissions on the
parent directories? Try:
ls -ld / /var /var/spool /var/spool/MIMEDefang
to see if any permissions are wrong.

drwxr-x--- 3 postfix postfix 4096 Jun 14 17:06 /var/spool/MIMEDefang
It is a problem from postfix (2.7.1) version and or mimedefang (2.69)
version?
