Here's what I usually I do when I want to create a new profile or new 
install:
1. backup all my relevant data
a. Mail directory (where all mail data is located)
b. News directory (where all news data is located)
c. abook.mab (the address book)
d. bookmarks.html (my bookmarks)

You can safely delete .msf files they will be recreated.

After all these are put in safe location, I create a new profile or let 
mozilla create a default profile on its first run. I then go to set my 
mail settings and afterwards I close mozilla including the quick launch. 
I then copy all my data to the appropriate directories overwriting as 
necessary. I then edit profile dependent data if necessary (open 
prefs.js and edit away, do this while mozilla is closed). =)
