Is the Harddisk ATA compliant?
Did you program it with the cartridge in slot 2 as the "readme"
discribes?
Do you use extra cartridges in your computer? -> pull all of them
out of your computer, except may be MSX-DOS 2.

This problem sounds familiar. It occured when I used a Megascsi
with it. Also the interface doesn't always like memorymappers.
