The mu portfile[1] lists --disable-mu4e as one of the configure
arguments, so unless you explicitly select the emacs variant of the mu
port, mu4e isn't installed at all. (I can't tell you how to select the
emacs variant in MacPorts, though... It's been a long time since I used
MacPorts, I preferred Homebrew when I still used OS X on my iMac.)

But when it's installed, you'll probably find mu4e in
/opt/local/share/emacs/site-lisp/ given that MacPorts installs stuff to
/opt/local.
