Yes it will work. I have used it in vmware for a couple of years now with no problems. However I have only scanned about 200 hosts with all but the DOS plugins enabled. The performance seemed to be fine I didn't notice a difference between running it natively or running it in a vmware instance. However it may depend on how much processing power you have available on your system.
