Just to be sure: c:/tmp and c:\temp are actually the same directory?

I can't help much with a MS Windows platform. What happens when you type
"c:\TeXLive\texmf-var\context\perltk\texfont.pl --help"? Is there some
special procedure for running perl programs on Windows?

Is the c:\TeXLive directory somehow write-protected or owned by another
user?

Do you get the same errors if you use "--fontroot=(some private
directory)", say "texmf" in your home directory?
