To my knowledge, there isn't a 64bit version of ESXi per say, it's just ESXi.
But, with the right hardware (aka modern CPU) and the correct BIOS settings, it will be able to run both 32and 64 bit guests at the same time. As far as backing up the VMs, once it's running, it's just a Windows box, so your BackupExec agents will work fine.
