Localizing your plates to some sort of SSD or RAID will speed up the interactivity as opposed to pulling frames over the network while you work. Also, setting up Nuke to work with half, or even quarter res jpg proxies will help for operations that don't involve color such as roto and tracking. There are also the Fusion IO cards which I've never worked with personally, but heard that they can provide great interactivity. The other option, of course, is to just run away and hide.
