we have been able to connect with Sourcefire Defense Centers 4 and 5. Which one are you using? IIRC, Sourcefire changed the licensing with 5.1 where you need to activate the interface explicitly via a license key. The error messages on the defense center don't give a helpful clue. You should check first the logs of the DC to see whether OpenVAS contacted the DC at all. Which version of the sourcefire plugin for OpenVAS are you using?
