A 320kbps mp3 file is good but not as good as a lossless format like wav, flac or lossless wma. You may only notice the short comings of mp3 when you play music back through a good hifi system. Of course, if you've ripped all your music to mp3 then it'll be too late to get that quality back unless you rip all over again.
I'm a firm believer that in the future everyone will use lossless music with very cheap storage and broadband being widespread and so I wouldn't want to back myself into that mp3 corner. It sounds great on portables and on the computer but lacks highs and lows, midrange detail and has a general boxed in kind of sound in comparison to lossless formats.
