Can an acrobat plugin written using Acrobat SDK 5.0 be used on a system with acrobat reader only and not writer.

Yes. PROVIDED that you get a Reader Integration Key from Adobe via their licensing process.

I have written a plugin but its not being detected by acrobat reader 5.0. What should be done to get it detected by acrobat reader???

First, go to http://partners.adobe.com/asn/tech/pdf/ikla.jsp and apply for a key.

After you've been approved, you'll sign a contract with Adobe and send them the required $$ as part of the contract. Upon reciept of the signed contract and the $$, Adobe will send you a Reader Integration Key.

You then use the tools provided in the Acrobat SDK in conjunction with yuor key to "Reader Enable" your plugin.
