Rob is correct. I use my D in the studio tethered to the computer, and all the images are transfered directly to the hard drive. I usually keep a card in the slot anyway, but I believe there is a special function that allows you to select "shutter release without CF card." I believe that the shutter won't release unless this is turned on.

The camera can work without a card, I've used it sans card. It can be tethered to a computer via the USB card and remote controlled using the Pentax Remote Assistant software. Probably not a bad way to go with it microscope mounted
actually.
