IMHO if you scan to 16 bit there's no need to adjust anything (except focus), because you are getting everything the sensor is capable of anyway. Since I don't have any software with 16 bit processing capability yet, it's better for me to adjust some controls on scanner...

Usually I set focus manually for each slide, then I turn on 16x multi pass scanning, and finally I adjust the dynamic range and the gray scale using the drivers controls.
