Did you set the K10D to save RAW files in DNG format? (Record menu, page 2, second item "Raw File Format" set to "DNG".)
The Lightroom v4.1 beta cannot open K10D PEF files. It opens K10D RAW/ DNG files without any problems. (Lightroom v1.0 release will open K10D PEF files too.)
