I don't know how to change the drive in Lightroom but assuming that 
you're using Microsoft WinXP you can semi-permanently assign a drive 
letter to your backup drive through the Computer Management Panel.  Open 
that application, (sorry I don't remember where XP has it hidden, select 
Storage->Disk Management from the Tree.  It will take a bit of time 
refreshing the display depending on your machines speed.  Select the 
device you normally store to and right click on it.  Select "Change 
Drive Letter and Path" from the drop down menu.  Edit that to be the 
drive letter expected by Lightroom.  Save and close everything. Next 
time you plug n a jump drive Windows should avoid assigning that drive 
letter to the jump drive.
