Yes, I think you're on the right thought process. Something is blocking the ARP broadcasts of router1 to router2. You can verify that with either tcpdump, snoop, or wireshark. If it's not too difficult, try replacing the switch and AP, one at a time, with a hub or a cable to bypass that device and definitely pinpoint the culprit.
