This looks correct and while I use Maildrop as my MDA here, I have one
setup where Postfix handles mail delivery to Maildirs on its own.

Just to check:
1. Are you sure there are no other conflicting home_mailbox lines
in main.cf that are uncommented?
2. Have you reloaded/restarted Postfix?
3. Have you used maildirmake to create a proper Maildir in your
user's home directory?
4. In /etc/login.defs have you replaced the MAIL_DIR and MAIL_FILE
directives with a single QMAIL_DIR directive with the value "Maildir"?
