"Automatically Detect Setting" on Microsoft Internet Explorer is NOT
what you want. Un-set that, it just adds useless overhead. Your browsers
should act as if there's no proxy in between. Your setup seems to be "by
the book" following both Squid FAQ-17[1] and the TransparentProxy
mini-HOWTO[2]. However I'm not sure if your gateway box (the one with
Squid installed) is already set as the gateway of your workstations.

Setting your gateway box as the gateway (aka default route) of your
workstations depends on how you set their networking up. If you set
networking up manually, then set this manually. If you use DHCP, you
want to set the "option routers" to the IP address of your
gateway/proxy.

This only means that Squid is set up correctly for manual proxy use.
When you have a manual proxy set and it is directly reachable, you don't
need a default route or gateway set up properly because your workstation
will know which machine to get in touch with. However with transparent
proxying set up, the transparent proxy must receive the packet and that
only happens when it is the gateway or default route.
