I don't know about locking the icons, but I dealt  with this problem 
by creating a quick and easy AppleScript application to reposition 
all my permanent icons (don't include icons that come and go from the 
desktop like CD icons, etc.):

First, make sure your icons are where you want them, then open the 
AppleScript Editor program. Then switch to the Finder, select all 
your permanent desktop icons or the group that you want, and drag 
them as a group to a slightly offset position. Next, switch to the 
Script Editor application and click the Record button. Switch back to 
the Finder by choosing it from the Application menu, which will 
ensure that the selected desktop icons remain selected, and drag the 
icons back to their former positions. Go back to the Script Editor 
once more, click the Stop button, and save the script as an 
application naming it to indicate the group of icons you are working 
with. Now you can reset your desktop icons by opening and running the 
application you just recreated.

Maybe this will work for your needs if you can't find information on 
locking them in place.
