domain1.com:mail1.domain1.com What is it mean?
mails for 'domain1.com' will be routed to mail1.domain1.com
I saw an example is: :mail1.mydomain.com
Is it mean all emails will route to mail1.mydomain.com?
If you add a rule like
:mail1.mydomain.com
All the mail from the GW1 is routed to mail1.mydomain.com, if you bounce a mail from GW1 it will go to mail1.
I suggest to install GW1 as a incoming and outgoing mailhub, to do this mail1 must not be reachble from the internet, otherwise you will receive some virus and spam directly to mail1. All the outgoing mail from mail1 can be routed to GW1 to send only clean mail.
In the gateway GW1, what should I install? 1. Qmail 2. Qmail-scanner 3. Anti-spam 4. Anti-virus
Is it enough? Do I need to install vpopmail?
In GW1 you don't need to install vpopmail.

In the GW1 add to the files:
/var/qmail/control/rcpthosts
domain1.com domain2.com domain3.com domain4.com
/var/qmail/control/smtproutes
domain1.com:mail1.domain1.com domain2.com:mail1.domain1.com # domain3.com:mail1.domain3.com domain4.com:mail1.domain4.com
You can Ip address or FQDN after the ':'
