Spamdyke needs to be on the perimeter (connected to the remote/sending server) in order to be effective. This is because most of what it does hinges on the remote/sending server's IP address, which is different for messages coming from the backup MX.

Some spammers target MX hosts with lower/lowest priority, as they sometimes don't have as strong of spam filtering in place.

So yes, you should by all means have spamdyke installed on backup MX servers.

The only reason I can think of to not have spamdyke installed would be for efficiency if it's not really needed. Even then though, I think that spamdyke's efficient enough that the difference between having it and not would be negligible.
