Yes, that is how I set mine up. If this was my box I would have it:
/home/rivendell-2/rivendell/music_101/*.mp3
or if your extensions are upper case
/home/rivendell-2/rivendell/music_101/*.MP3

or if you have a mix of upper and lower case extensions, have two dropboxes
