Not much to add except to take advantage of the VM and take a snapshot before you do the upgrade. You don't say where your database is stored but even so, you will be looking at a DB restore only if things go a bit wrong. I bought a license for BPS - BES and a support contract. The support contract is supposed to include a free BES 5 upgrade. Can I go from BPS 4 - BES 5, or do I need to go BPS 4 - BES 4 - BES 5. Once D: is added, you can change the logfiles so that they output to D: instead of C: that alone will give C: plenty of breathing room. If your server is 2008 or 2008 R2 it's even easier, expand drive in vSphere client than in Windows go to disk management, right click the drive and expand. No rebooting or downtime required.
